Yes, you can define your own roles and define role action for the created role.
One employee can have multiple roles, but only one designation.
If the designation is not used anywhere then you can delete it.
You can simply put in a dummy email id while creating employee record for those users.
Any given employee will have a lot of records associated with his/her name. So deletion is difficult, but you can mark the employee as ‘Resigned’ and it will not show up anywhere in the system.
As of now, one employee can report only to one manager.
